In the context of climate change, an increased frequency of drought stresses might occur at a regional scale in boreal forests. To assess photosynthetic responses to drought treatment, seedlings of 12 open-pollinated families of white spruce (Picea glauca (Moench) Voss) differing in their growth performance were grown in a controlled environment. Gas exchange and chlorophyll fluorescence parameters as well as shoot xylem water potential (WP) were measured for 21 successive days after watering was stopped. Net photosynthesis decreased as stomatal conductance decreased. Net photosynthesis was not affected by drought until WP reached –2.0 MPa when stomata were closed. Initial fluorescence (F and basic fluorescence after induction (F00) were not affected by drought. A progressive decrease in maximal (Fm) and variable fluorescences (Fv), maximum photosystem II (PS II) efficiency (Fv = Fm), effective quantum yield of PS II (FII), photochemical efficiency of open PS II (Fp), and photochemical quenching (qP) was observed at WP < - 1.0 MPa, whereas non-photochemical quenching (qN) remained high throughout the drought treatment. White spruce families with inferior growth performance showed higher values of Fm, Fv, Fv = Fm, Fp, and qN at WP< - 2.0MPa. The results indicated that chlorophyll fluorescence variables can be used as drought markers in relation to present or predicted climate conditions. These could be used for selecting planting stock adapted to drought periods or dry environments. These markers showed that slow-growing genotypes are better adapted to drought conditions than intermediate or fast-growing genotypes in present and predicted drought conditions.  相似文献

The efficacy of the organic insecticide Diatect II against boll weevil (Anthonomus grandis Boheman) in cotton (Gossypium hirsutum L) in the Lower Rio Grande Valley of Texas were assessed in small-plot field trials and greenhouse cage tests using azinphos-methyl treatments as a standard for comparison. Plastic sheets were placed in the furrows of the treated plots to retrieve boll weevils which dropped from the plants after being killed by the insecticides. Samples of live weevils taken by a tractor-mounted vacuum sampler revealed a modest, but significant, reduction in boll weevil populations in Diatect II plots. However, samples of dead weevils indicated that this reduction was due to movement of weevils out of the plots rather than to mortality. This interpretation is supported by greenhouse cage studies, where mortality in Diatect II treated cages was no greater than that in untreated control cages. The effects of insecticide treatments in small plots can be confounded easily and quickly by interplot movement of target insects. Although the relative effects of various compounds can usually be assessed by sampling the populations in plots soon after treatment, the best measure of efficacy is obtained by directly sampling insects that have died in the plot. This parameter is insulated from the effects of interplot movement, unless the toxicant is slow to immobilize the target insect. Taken together, our results indicate little efficacy by Diatect II against boll weevil under our test conditions.  相似文献

The main mode of herbicidal activity of 2-hydroxy-3-alkyl-1,4-naphthoquinones is shown to be inhibition of photosystem II (PSII). The herbicidal and in vitro activities have been measured and correlated with their (Log)octanol/water partition coefficients (Log Ko/w). The length of the 3-n-alkyl substituent for optimal activity differed between herbicidal and in vitro activity. The maximum in vitro activity was given by the nonyl to dodecyl homologues (Log Ko/w between 6.54 and 8.12), whereas herbicidal activity peaked with the n-hexyl compound (Log Ko/w = 4.95). The effect of chain branching was also investigated using isomeric pentyl analogues substituted at position 3. All exhibited similar levels of in vitro activities but herbicidal activities differed, albeit moderately, with the exception of one analogue that was much less phytotoxic. Other modes of action were also investigated using two representative compounds. They did not show any activity on photosystem I or mitochondrial complex I, or generate toxic oxygen radicals by redox cycling reactions. Only moderate activity was found against mitochondrial complex III from plants, in contrast to much higher corresponding activity using an insect enzyme.  相似文献

选用128头健康“杜长大”三元杂交断奶仔猪,随机分为对照组、CuSO4组、纳米蒙脱石(MMT)组和MMT-Cu组,分别饲喂基础日粮(对照组)、基础日粮+78 mg/kg Cu2+(CuSO4.5H2O,相当于MMT-Cu中Cu2+的量)、基础日粮+2.0 g/kgMMT、基础日粮+2.0 g/kgMMT-Cu。结果表明,与对照组相比,MMT-Cu组能显著提高(P<0.05)断奶仔猪的平均日增重、饲料转化效率及小肠绒毛高度、绒毛高度与隐窝深度的比值,显著降低(P<0.05)腹泻率及小肠、结肠内容物中大肠杆菌和沙门氏菌的数量;而MMT组也显著降低(P<0.05)仔猪腹泻率,提高(P<0.05)小肠绒毛高度和绒毛高度与隐窝深度的比值。与MMT组或CuSO4组相比,MMT-Cu组能显著(P<0.05)提高断奶仔猪的平均日增重和饲料转化效率(P<0.05),显著降低了(P<0.05)小肠和结肠内容物中大肠杆菌和沙门氏菌的数量。  相似文献

OBJECTIVE: To compare the expression of major histocompatibility complex (MHC) class II antigen in the corneas of normal dogs and dogs affected with chronic superficial keratitis (CSK). METHODS: MHC class II expression was determined in frozen sections of normal canine cornea and cornea from lesions of CSK by immunohistochemistry using a monoclonal antibody directed against the canine MHC class II molecule. Langerhans cell phenotype was determined morphologically and by histochemical determination of ATPase activity. To determine the influence of gamma interferon on expression of MHC class II molecules by corneal cells, corneal explants were cultured with the cytokine and MHC class II expression determined as above. RESULTS: Numerous MHC class II-expressing cells were demonstrated within the stroma and epithelium of the normal corneal limbus and conjunctival epithelium while very little MHC class II expression was detected in the central region of normal canine cornea. In limbal and conjunctival epithelium, cells expressing MHC class II antigen showed ATPase activity, suggesting that they were Langerhans cells. Corneas from dogs with CSK showed MHC class II expression associated with stromal cells, some of which exhibited a dendritic morphology while most were lymphocytic. Corneal epithelial cells within the lesion also aberrantly expressed MHC class II. Corneal explants expressed MHC class II to varying degrees after differing periods of incubation with the cytokine gamma interferon. CONCLUSIONS: While the normal central cornea has little MHC class II expression, aberrant expression occurs in CSK, associated with secretion of gamma interferon by infiltrating CD4-expressing lymphocytes. Although this change is likely to be a secondary feature of the CSK lesion, increased MHC class II expression may play a part in perpetuating the corneal inflammation seen in the disease.  相似文献

The aim of this study was to characterize the phenotype of leukocytes infiltrating the duodenal mucosa of cats with inflammatory bowel disease (IBD) by using immunohistochemistry and computer-aided morphometry to assess whether immunologic markers would aid in characterization of IBD. Frozen and formalin-fixed duodenal biopsies were collected from cats referred for investigation of chronic vomiting, diarrhea, or both (n = 34). Reference ranges were previously established by using duodenal samples from healthy cats (n = 16). No significant difference was found in the number of immunoglobulin G+ (IgG+) or IgA+ in either the villous lamina propria or the crypt lamina propria between cats with IBD and control cats. T cells (CD3+) increased in number from crypt to the tip of the villi in biopsies from both diseased (mean +/- SD for each group was 18.8 +/- 6.6 and 17.7 +/- 4.2 cells/ 10,000 m2 in cryptal areas to 25.2 +/- 9.5 and 29.1 +/- 13.3 cells/10,000m2 in villous areas) and healthy animals (17.9 +/- 3.9 cells/10,000 microm2 in cryptal areas to 24.1 +/- 9.3 cells/10,000 microm2 in villous areas) and no significant difference was found between diseased and control cats. By contrast, major histocompatibility complex (MHC) class II expression by leukocytes with dendritic cell or macrophage morphology in the lamina propria was significantly greater in cats with IBD (13.3 +/- 4.2 cells/10,000 microm2 in cryptal area; P = .016) than in healthy cats (11.9 +/- 3.0 cells/10,000 microm2) and MHC class II expression by enterocytes also was more pronounced in these cats showing an overall intensity of expression of 7.1 +/- 4.0 cells/10,000 microm2 in cats with IBD as opposed to 0.0 +/- 0.0 cells/10,000 microm2 to 0.3 +/- 0.7 cells/10,000 microm2 in healthy cats. These findings suggest that a subtle immunologic dysregulation occurs in spontaneously arising feline IBD.  相似文献
